My profile is: Employed full time, no business, no self employment, just T4 . Simple.
Now, last year I invested money in a real estate project for which I got upfront interest. Now the company issued me T4A with the box 20 filled with the upfront interest I received.
I understand that T4A is issued as commission income to self employed people. Does that means I need to fill T2125 for the business expenses occurred? Does that also means I need to file GST/HST return?

Box 20 of a T4A slip is "Self-employed commissions".
These are the instructions on the back of the slip for Box 20:
Box 020, Self-employed commissions – Enter your gross commissions income on line 13899 and your net commissions income on line 13900.
All that doesn't sound like interest.
Are you sure that was interest you were paid? You may have invested directly without an agent and that was actually the sales commission for selling the investment to yourself!

Interest in not reported in Box 20 of a T4A, issuer is indicating this is not interest.
Re OP's questions:
I'm no expert but I notice H&R Block site indicates "Amounts in box 20 (self-employment commissions)……….. of a T4A slip are treated as self-employment income. If you have an amount in box 20...….., you’ll need to also report this income and any related expenses on the T2125: Statement of business or professional activities page in H&R Block’s tax software."
Re GST/HST, you're only required to file a return if you're required to collect GST/HST and in this case that would depend on when the $30K registration threshold was exceeded, if it was.
